Seventy nine percent of dose /about 50 mg/kg sc admin to guinea pigs/ was eliminated as thiocyanate in urine. ... Skin penetration was suggested by incr in thiocyanate in urine ... after application to depilated skin. Greater quantities were absorbed when skin was abraded.

The concentrations of thiocyanate in the blood and urine of guinea pigs injected with adiponitrile were proportional to the doses administered. Following administration of adiponitrile, 79% was eliminated as thiocyanate in the urine of guinea pigs. Of the cyanide antidotes, thiosulfate was most effective. However, on the basis of the ratio between administered adiponitrile dose and quantity of cyanide detected, /it was/ concluded that a greater part of the dose was metabolized to cyanide.

... In guinea pigs ... it is hydrolyzed to hydrogen cyanide in the body giving rise to thiocyanate in urine.

Male ddY mice were pretreated with carbon tetrachloride or olive oil and administered adiponitrile orally in concentrations sufficient to determine its median lethal dose (LD50). Brain cyanide concentrations were determined. The distribution coefficient (P) for partitioning adiponitrile between water and n-octanol was measured. Mice were pretreated as before and dosed with adiponitrile at 3 to 5 times its LD50. Mean survival times were determined. Hepatic microsomes were obtained from mice pretreated with carbon tetrachloride or olive oil and incubated with adiponitrile. The rate of release of free cyanide was investigated. The oral LD50 for animals pretreated with olive oil was 1.592 mmol/kg. The LD50 for the carbon tetrachloride treated mice was 2.652 mmol/kg. Brain cyanide concentrations in olive oil pretreated mice ranged from 0.35 to 0.74 ug/g. Carbon tetrachloride pretreatment lengthened survival and reduced brain cyanide concentrations. Dinitriles are metabolized to cyanides in vivo and in vitro.

PharmaCompass offers a list of Adiponitrile API manufacturers, exporters & distributors, which can be sorted by GMP, USDMF, JDMF, KDMF, CEP (COS), WC, Price, and more, enabling you to easily find the right Adiponitrile manufacturer or Adiponitrile supplier for your needs.
Send us enquiries for free, and we will assist you in establishing a direct connection with your preferred Adiponitrile manufacturer or Adiponitrile supplier.
PharmaCompass also assists you with knowing the Adiponitrile API Price utilized in the formulation of products. Adiponitrile API Price is not always fixed or binding as the Adiponitrile Price is obtained through a variety of data sources. The Adiponitrile Price can also vary due to multiple factors, including market conditions, regulatory modifications, or negotiated pricing deals.
A Adiponitrile manufacturer is defined as any person or entity involved in the manufacture, preparation, processing, compounding or propagation of Adiponitrile, including repackagers and relabelers. The FDA regulates Adiponitrile manufacturers to ensure that their products comply with relevant laws and regulations and are safe and effective to use. Adiponitrile API Manufacturers are required to adhere to Good Manufacturing Practices (GMP) to ensure that their products are consistently manufactured to meet established quality criteria.
A Adiponitrile supplier is an individual or a company that provides Adiponitrile active pharmaceutical ingredient (API) or Adiponitrile finished formulations upon request. The Adiponitrile suppliers may include Adiponitrile API manufacturers, exporters, distributors and traders.
Adiponitrile Active pharmaceutical ingredient (API) is produced in GMP-certified manufacturing facility.
GMP stands for Good Manufacturing Practices, which is a system used in the pharmaceutical industry to make sure that goods are regularly produced and monitored in accordance with quality standards. The FDA’s current Good Manufacturing Practices requirements are referred to as cGMP or current GMP which indicates that the company follows the most recent GMP specifications. The World Health Organization (WHO) has its own set of GMP guidelines, called the WHO GMP. Different countries can also set their own guidelines for GMP like China (Chinese GMP) or the EU (EU GMP).
PharmaCompass offers a list of Adiponitrile GMP manufacturers, exporters & distributors, which can be sorted by USDMF, JDMF, KDMF, CEP (COS), WC, API price, and more, enabling you to easily find the right Adiponitrile GMP manufacturer or Adiponitrile GMP API supplier for your needs.
A Adiponitrile CoA (Certificate of Analysis) is a formal document that attests to Adiponitrile's compliance with Adiponitrile specifications and serves as a tool for batch-level quality control.
Adiponitrile CoA mostly includes findings from lab analyses of a specific batch. For each Adiponitrile CoA document that a company creates, the USFDA specifies specific requirements, such as supplier information, material identification, transportation data, evidence of conformity and signature data.
Adiponitrile may be tested according to a variety of international standards, such as European Pharmacopoeia (Adiponitrile EP), Adiponitrile JP (Japanese Pharmacopeia) and the US Pharmacopoeia (Adiponitrile USP).
